Abstract: | The intestinal microbiota encompasses the inhabitant intestinal micro-organisms among which are included lactobacilli, facultative anaerobic bacteria (aerobes) and bifidobacteria. Microbiota has a critical role in intestinal homeostasis that can be disrupted by stress. The latter triggers the release of catecholamines by the nerve fibers of sympathetic nervous system (SNS) with effects on intestinal permeability and inflammation. Justification: Outcome of stress through the SNS on the intestinal microbiota under conditions of treatment with modulating proteins such as bovine lactoferrin (bLf) are not known. Aim: To evaluate the participation of the SNS on the fecal bacterial count of mice subjected to stress and treated with bLf. Methodology: Groups of 8-week age female BALB/c mice treated with bLf 0.1% w/v for 7 days in the drinking bottle were subjected to board-immobilization stress for 2h/4days (n=10, bLf-st) or no stress (n=10, bLf-unst); Other groups of mice treated with water in drinking bottle underwent stress (n=10, ctl-st) and no stress (n=10, ctl-unst) were included. Each group was subdivided in groups of 5 mice each treated intraperitoneally with the sympathetic neurotoxin DSP4 (bLf-st-DSP4, bLf-unst-DSP4, water-st-DSP4 and water-unst-DSP4) or with isotonic saline as vehicle (bLf-st-Vh, bLf-unst-Vh, water-st-Vh and water-unst-Vh). Fecal samples were collected and cultured at 37°C either aerobically on TSA agar (24 h, aerobes) or anaerobically on MRS agar (48 h, lactobacilli) and TOS agar (72 h, bifidobacteria). Colonies were counted and data were expressed as colony-forming units per g of feces (CFU/g). Results: Lactobacilli count in the bLf-st-DSP4 group was significantly lower (P<0.05) in relation to the water-St-Vh group. No other significant difference in bacterial count was observed. Conclusion: These findings indicate that stress did not display actions on the bacterial count through the SNS. However, data suggested that bLf favored the participation of the SNS on the reduction of fecal lactobacilli count under stress conditions. La microbiota intestinal es el conjunto de micro-organismos que residen en el intestino, entre los cuales se encuentran lactobacilos, bacterias anaerobias facultativas (aerobias) y bifidobacterias. La microbiota tiene un papel importante en la homeostasis intestinal que puede verse afectada por el estrés. Este último, desencadena la liberación de catecolaminas por parte del Sistema Nervioso Simpático (SNS) con efectos sobre la permeabilidad intestinal y la inflamación. Justificación:. Los efectos de las catecolaminas liberadas por el SNS en la microbiota intestinal no han sido evaluadas en condiciones de estrés y bajo el tratamiento con bLf Objetivo: Evaluar la participación del SNS sobre la cuenta bacteriana fecal de ratones sometidos a estrés y tratados con bLf. Metodología: Grupos de ratones hembras BALB/c de 8 semanas tratados con bLf 0.1% p/v durante 7 días en el bebedero (n=20) fueron sometidos a estrés por inmovilización en tablero 2h/4dias (n=10 bLf-st) o sin estrés (n=10 bLf-unst); se incluyeron otros grupos de ratones tratados con agua (n=20) en el bebedero con estrés (n=10 ctl-st) y sin estrés (n=10 ctl-unst). Cada grupo se subdividió en grupos (n=5) tratados vía intraperitoneal con la neurotoxina simpatética DSP4 (bLf-st-DSP4, bLf-unst-DSP4, agua-st-DSP4 y agua-unst-DSP4) o con salina isotónica como vehículo (bLf-st-Vh, bLf-unst-Vh, agua-st-Vh y agua-unst-Vh). Se recolectaron heces de los ratones para su cultivo a 37°C en agar TSA en aerobiosis (24 h, aerobios) y en anaerobiosis en agar MRS (48 h, lactobacilos) y agar TOS (72 h, bífidos). Se realizó la cuenta total de colonias y los datos fueron expresados en unidades formadoras de colonias por g de heces (UFC/g). Resultados: Se observó que la cuenta de lactobacilos en el grupo bLf-st-DSP4 fue significativamente menor (P<0.05) en relación al grupo agua-St-Vh. No se observó alguna otra diferencia significativa en la cuenta bacteriana. Conclusión: Los resultados indican que no hubo efecto del estrés por sí mismo sobre la cuenta bacteriana a través del SNS. Sin embargo, los datos sugieren que la bLf favorece la participación del SNS sobre la reducción de lactobacilos en condiciones de estrés. |
